fa بررسی فیلوژنی مولکولی شقایق دریایی جنس Actiniaria: Actiniidae)Anemonia بر اساس توالی DNA میتوکندریایی (COI) در خلیج فارس پژوهشي Research/ Original/ Regular Article <p style="text-align: justify;"><span dir="RTL">در این مقاله، فیلوژنی مولکولی شقایق دریایی جنس (</span><span dir="RTL">Actiniaria: Actiniidae</span><span dir="RTL">) بر اساس توالی D‏NA&nbsp;</span><span dir="RTL"> میتوکندریایی (</span><span dir="RTL">COI) در خلیج فارس مورد بررسی قرار گرفت. گونه مورد مطالعه در این تحقیق از جزیره لارک واقع در خلیج فارس جمع آوری شد وDNA&nbsp;</span><span dir="RTL"> آن استخراج و قسمتی از ژنCOI&nbsp;</span><span dir="RTL"> آن (حدود 700 جفت باز) با استفاده از دو پرایمرLCO1490</span>&nbsp; <span dir="RTL">وHCO2198&nbsp;</span><span dir="RTL"> به وسیلهPCR</span>&nbsp; <span dir="RTL">تکثیر داده شد و محصولPCR&nbsp;</span><span dir="RTL"> در دستگاه توالی یاب خودکارDNA&nbsp;</span><span dir="RTL"> به وسیله شرکتBioNEER&nbsp;</span><span dir="RTL"> توالی&shy; یابی گردید. توالی نوکلئیک اسید و پروتئین گرفته شده از ژنCOI&nbsp;</span><span dir="RTL"> به عنوان یک توالی جدید در ژن بانک گزارش شد. بر طبق این توالی شقایق خلیج فارس به عنوان یک گونه جدید با نامAnemonia sp. PG&nbsp;</span><span dir="RTL"> و شماره ثبتAB933643.1&nbsp;</span><span dir="RTL"> در وب&shy; گاهNCBI&nbsp;</span><span dir="RTL"> قرار گرفت. هدف از این مطالعه شناسایی مولکولی شقایق خلیج فارس و انجام آنالیز مولکولی روی آن بود. در آنالیز مولکولی توالی قسمتی از ژنCOI&nbsp;</span><span dir="RTL"> میتوکندریایی گونهAnemonia sp. PG&nbsp;</span><span dir="RTL"> از خلیج فارس با 15 شقایق دریایی دیگر از بانک ژن که از لحاظ توالی ژنتیکی به گونه مورد بررسی نزدیک هستند مقایسه گردید. گونهMetridium senile&nbsp;</span><span dir="RTL"> به عنوان برون گونه در نظر گرفته شد. بررسی فیلوژنتیکی بر اساس آنالیزNeighbor-Joining&nbsp;</span><span dir="RTL"> رابطه خواهری بین گونهAnemonia sp. PG </span>&nbsp; <span dir="RTL">از ایران و گونهAnemonia sp. Anem&nbsp;</span><span dir="RTL"> را نشان داد ولی روابط منوفایلتیک بین خانواده‌های شقایق دریایی را نشان نداد.</span></p> <p style="text-align: justify;">Sea anemones (order: Actiniaria) are a subclass of Hexacorallia. They are very varied and are organisms with a simple skeletal polyp. The specie in this study was collected from Larak Island in the Persian Gulf. DNA was extracted from the sample and a part of mtDNA COI gene was amplified. Two pairs of primers were designed to amplify a final target of 600 bp using PCR method which the product was successively sequenced. The purpose of this study was the molecular identification of the Persian Gulf sea anemone. The nucleotide and protein sequences obtained from this anemone&rsquo;s COI gene were annotated in NCBI Gene Bank as <em>Anemonia</em> sp. PG, the PG standing for the Persian Gulf.&nbsp; Phylogenetic studies were performed to compare the partial sequences of COI genes of 16 sea anemones (order: Actiniaria) with the newly identified anemone (<em>Anemonia</em> sp. PG).&nbsp; Phylogenetic studies based on Neighbor-Joining analysis showed sister relations between <em>Anemonia</em> sp. PG species from Iran and <em>Anemonia</em> sp.
