fa بررسی ریخت‌شناسی و شناسایی مولکولی گونه Solen dactylus (Cosel, 1989) از خور گوبان، شمال خلیج‌فارس Morphological investigation and molecular identification of Solen dactylus (Bivalvia: Solenidae) in the Khowr-e Guban (North of Persian Gulf) زيست شناسی دريا Marine Biology پژوهشي Research/ Original/ Regular Article <div style="text-align: justify;">پیشینه و اهداف: هدف از این تحقیق بررسی ریخت‌شناسی و شناسایی مولکولی صدف دسته‌چاقویی Solen dactylus میباشد. گونه‌های مختلفSolen spp.  به عنوان صدف دسته چاقویی یا razor clam  شناخته می‌شوند. این صدف به‌طور گسترده در بازارهای بین‌المللی به عنوان کالا و غذا مصرف میشود و تحقیقات بسیاری در مورد آن انجام شده است. روش‌ها‌: نمونه‌برداری این گونه از ایستگاه‌های تعیین شده در خور گوبان واقع در جنوب تالاب شادگان در دو فصل زمستان و تابستان سال 1396 انجام شد. شناسایی ریخت‌شناسی و با استفاده از کلیدهای شناسایی معتبر صورت گرفت. همچنین شناسایی مولکولی این گونه با استفاده از تکثیر قطعات ژنومی ناحیه COI ژنوم میتوکندی انجام شد. توالی ژنومی حاصل با استفاده از نرم‌افزارهای Chromas ، BioEdit  و Mega 7  مورد تجزیه وتحلیل قرار گرفت. همچنین بر اساس مقایسه نتایج توالی با سایر داده های موجود در NCBI  درخت فیلوژنی ساخته شد. یافته‌ها: نتایج نشان داد که گونهی  Solen strictus ، با گونهی Solen marginatus   و گونه‌ی تحقیق حاضر (Solen sp.) در یک کلاد خواهری  قرار می‌گیرند و از یک دنیای مشترک انشقاق یافته‌اند. توالی قطعات ژنی تکثیر یافته با پرایمرهای اختصاصی COI، شباهت 83 درصدی را با گونه‌ی S. strictus نشان داد. این در حالی است که این دو گونه از لحاظ شباهت ظاهری به یکدیگر شبیه‌اند، اما در بررسی‌های ریخت‌شناسی، وجود مشخصاتی چون تانتاکول‌های پالیالی در قسمت جلویی پا، وجود دندانه‌ی کوچک، اندازه‌گیری ابعاد طول و عرض صدف که ویژگی شاخص گونه‌ی S. dactylus است و در گونه‌های S. strictus و S. marginatus وجود ندارد، مشاهده گردید. نتیجه‌گیری: بنابراین با توجه با نتایج ریخت‌شناسی و مولکولی به نظر می‌رسد گونه Solensp.  همان گونه S. dactylus است. این گونه در پایگاه اطلاعاتی NCBI  با کد پذیرش QGL52628  ثبت گردید.</div> <div style="text-align: justify;">Background and Objectives: The aim of this study was to investigate the morphology and molecular identification of oyster knife Solen dactylus. Razor clams are edible bivalve molluscs that fetch high prices in international markets.</div> <div style="text-align: justify;">Methods: For this purpose, sampling of this species was performed from the designated station in Khowr-e Guban, south of Shadegan wetland. Morphological identification was first performed using valid identification keys. Then, to ensure assurance, molecular identification of the species was performed using amplification of mitochondrial COI region. The resulting genomic sequences were analyzed using Chromas, BioEdit, and Mega 7 software. Blast sequences were also identified in NCBI and phylogenetic tree was constructed. Findings: The results showed that the Solen strictus species and the Solen marginatus species and the present study species (Solen sp.) are in a sister clade and derived from a common ancestor. The sequence of amplified gene fragments with specific COI primers showed 83% similarity to S. strictus, while the two species are similar in appearance, but in morphological studies and the use of identification keys specifications such as palliative tentacles on the front of the foot, small teeth, and measurement of oyster length and width, which are characteristic of S. dactylus and do not exist in S. strictus and S. marginatus species. Conclusion: Therefore, according to morphological and molecular results, Solen sp. species is probably the same as S. dactylus.
